Detached and stoic, Amado speaks his mind bluntly no matter what others make of him, and he keeps up a habit of smoking. Though outwardly respectful, he holds much the same low regard for others' lives as Jigen when a goal is at stake, yet he leans practical and dislikes needless waste, abandoning any project that stops showing progress. Almost all of this grows from a heart broken by his daughter's death and his failure to save her; everything since has served the aim of bringing her back. He rarely loses his composure, having salted any situation with enough leverage to keep the upper hand, and he is deeply manipulative, fond of manufacturing a crisis only he can resolve. He openly admits he lacks any iron will, could not withstand torture, and will beg for his life without shame, seeing no worth in pride when survival is on the line.

At first his loyalty to Kara ran no deeper than self-interest, since he expected the organization to revive Akebi as his reward. Once he understood that Isshiki's design would consume the entire planet, and doubted the bargain would ever be honored, he broke faith and set himself against the whole Otsutsuki clan, judging their history parasitic and vile. Even after siding with Konoha he never stopped scheming, quietly reapplying a Kama to Kawaki without approval and hiding a shutdown command inside it. He carries a dry, twisted humor as well, joking that killing Kawaki was one way to block Isshiki's return even as he had already made sure it would not come to that.

Amado's daughter Akebi died of an unexplained illness roughly twelve years before Isshiki Otsutsuki's assault on Konoha. Rather than chase a cure, he tried to resurrect her by cloning her body and transferring her preserved memories, but the clones he produced, later called Delta, shared her face and recollections while remaining entirely different people. His repeated failures broke him, and it was then that Jigen recruited him with a promise to restore his daughter. Within Kara he rose to Inner and head of research and development, outfitting the members with Scientific Ninja Tools, grafting Shibai Otsutsuki's DNA into them to awaken shinjutsu, and building cyborgs such as Ada and Daemon whose power outstripped Jigen's. He also cloned Jiraiya to create Koji Kashin as a secret ally and extensively rebuilt Kawaki into a living weapon.

An unmatched engineer, Amado is credited with Kara's entire arsenal of Shinobi-Ware and autonomous puppets, and even Konoha's own genius Katasuke Tono conceded his talents ran well past his own. Kawaki called him a technological genius, and Isshiki himself sought him out. His grasp of the Kama later let him restore Kawaki's mark after Isshiki's soul was extinguished, and he devised a drug to weaken the Byakugan as a weapon against the Otsutsuki. Once he grasped that Isshiki intended to end all life, he conspired with Koji to kill Jigen and forced Isshiki into reincarnating in an imperfect vessel, a maneuver that stripped away Kawaki's Kama and left the enemy open to a permanent death.

Fleeing Kara, Amado defected to Konoha, taking Shikadai hostage with a bomb collar he later revealed to be a fake, and traded all of his intelligence on Kara, Jigen, and the Otsutsuki for citizenship and protection. He was granted asylum under strict conditions and allowed to work with the Scientific Ninja Weapons Team. Still fixed on his daughter, he secretly branded Kawaki with a fresh Kama seeded with Akebi's data, planning to place that mark on a cloned body. After the Omnipotence incident scrambled the world's memories and drove Boruto from the village, Amado eventually noticed the discrepancies between his recollections and reality, realizing Kawaki, not Boruto, was the one he needed. In the later conflict against the sentient God Trees, Kawaki forced him to raise his combat output to maximum, and Amado agreed to boost his power in exchange for eventually reviving Akebi, at one point rigging an engineered toad to switch Kawaki's settings remotely and later using it to recapture the surviving Koji.
