DIO's Bone is a fragment of DIO's own body, handed to the priest Enrico Pucci as a keepsake before the events of Stone Ocean. Seemingly a simple relic, it becomes the grotesque seed from which the Green Baby, and ultimately the Stand C-Moon, is born.
Judging by its contour and the two rounded knobs on it, the so-called sesamoid bones, the relic is DIO's first metatarsal, one of the bones seated in the foot just behind the big toe. Pucci keeps it close as a memento long before it shows any strange properties.
The bone changes everything once Pucci turns Sports Maxx's Stand, Limp Bizkit, on it to restore it to life, whereupon it turns contagious. Anyone who lays a hand on it begins to mutate, slowly sprouting until the body becomes a kind of plant. Those transformed convicts, thirty-six of them drawn from Green Dolphin Street Prison, drink in sunlight and feed the gathered nourishment to an infant-like being often called DIO's homunculus, better known as the Green Baby. At its birth a shard of DIO's soul is reborn inside it. The creature then seeks Pucci out and merges with his Stand, giving rise to C-Moon.
Enrico Pucci is the bone's keeper, receiving it in 1988 after DIO, uneasy that Whitesnake could steal The World at will, tested his loyalty; Pucci swore he loved DIO as he loved God, and DIO tore the bone from his body in apology. In truth the bone comes from Jonathan Joestar, whose body DIO claimed at the close of Phantom Blood, and it is through this link that the Green Baby, Pucci, and his twin Weather Report all bear the Star Birthmark.

Despite the name, DIO's Bone actually comes from Jonathan Joestar, whose body DIO took over at the close of Phantom Blood. This link is why the Green Baby, Pucci, and his twin Weather Report all carry the Star Birthmark.
