Penelope has pink hair worn short, and she favors sun dresses along with lipstick and nail polish. The novel's interior illustrations render her as blonde, while the covers keep her hair pink.

On the job at Erina's company she puts on glasses, ties her hair up, and wears a knee-length dress, and Jorge considers her beautiful.

Once trapped in an abusive relationship with a man named Edvard, Penelope stayed loyal to him despite his cruelty, and the terror inflicted on her in her dreams by a serial killer twisted her fear of clowns into a full phobia and a Wound. Emotionally fragile at first, she steadies and gains confidence after moving in with the Joestars, becoming a kind, sisterly presence to Jorge.

That gentleness has limits: she turns aggressive and nearly kills Edvard on seeing him again, and she is fiercely protective, threatening bullies who target Jorge and standing by him even when he is accused of murder. She and Erina rejoice together at news of Elizabeth's pregnancy and her marriage to Jorge.

Penelope's power is a Wound she describes as a clown, surfacing only when she is stressed or afraid; it conjures locked rooms and hangs its victims inside. Without realizing it she also develops a Stand, Cube House, which is in truth Reimi Sugimoto's main body.

Her parents divorced around 1900, and though her mother won custody, Penelope blamed her for driving her father away. Edvard abused her, sold off her belongings, and threatened her, and it was then that clowns began invading her dreams. She crosses paths with the young detective Jorge and his partner Tsukumojuku Kato as they investigate a string of locked-room murders, of which she is one victim, and she helps them expose the culprit, a fifteen-year-old named Javier Cortez.

In 1904, forced from her home, she is taken in by Erina and works at the La Palma branch of the Star Mark Trading company, tutoring Jorge and treating him as a younger brother, until her ex resurfaces and she nearly kills him for his past cruelty. Following Jorge back to his own universe, she marries his adoptive father Jonda Joestar, and a year later the couple have a daughter, leaving her known afterward as Penelope Joestar.
